Sildenafil Increases Serum Testosterone Levels by a Direct Action on the Testes

Matthew Johnathan Spitzer, MD1, Shalender Bhasin, MD1, Thomas G Travison, PhD1, Maithili Davda, MPH1, Helene Stroh, BA1 and Shehzad Sultan Basaria, MD MBBS11 Boston University Medical Center, Boston,MA
Phosphodiesterase-5-inhibitors, such as sildenafil, increase intracavernosal cyclic guanosine monophosphate levels, which results in corporal vasodilatation and induces penile erection. Administration of sildenafil to male rats has been shown to increase serum testosterone levels by directly stimulating testicular Leydig cells. However, the effect of sildenafil on the hypothalamic-pituitary-gonadal axis in humans has not been evaluated in the setting of a large clinical trial. The Testosterone and Erectile Dysfunction trial (ClinicalTrials.gov # NCT00512707 ) administered an optimized dose of sildenafil in an initial run-in period to 140 men, age 40-70 years with erectile dysfunction and low testosterone levels (< 330ng/dL) and then randomized them to either transdermal testosterone or placebo gel. Serum testosterone, dihydrotestosterone and estrogens were measured using LC-MS/MS.

Administration of an optimized dose of sildenafil was associated with mean increases of 103ng/dL (P < 0.001) and 60.1pg/mL (P < 0.001) in total and free testosterone levels, respectively. This was accompanied by parallel increases in serum dihydrotestosterone (48.8pg/mL; P < 0.001), estradiol (3.7pg/mL; P < 0.001), and estrone (2.9pg/mL; P = 0.012) levels, and significant suppression of luteinizing hormone (change -1.3units/L; P = 0.003) levels, suggesting a direct effect at the testicular level. FSH and inhibin B levels remained unchanged.

Conclusion: Sildenafil administration was associated with increased testosterone levels likely due to a direct effect on the testis.

Amazing! I tell everyone I know that all men over the age of 25 should take a low dose of Cialis daily. I firmly believe this. It has shown, and proven to help with prostate issues. It is also being commonly prescribed to people for BP issues. Want to experiment and have some fun? Give your lady 5-10mgs of it every now and then. If I were honest with you guys, if someone gave me the option of AAS's or Cialis, I'd choose Cialis 100% of the time. Not for the erections, that is just a plus. It is for the overall well being. I feel good, my BP is better controlled, and vascularity is second to none. And don't get me started about the "pumps" at the gym! WOW!

Admin, it really is amazing for ED use, at appropriate dosages. I cant say I have seen long term studies, but I have spoken to several MDs, and every one of them said the same thing. They all seem to be on board with Cialis (not Viagra) due to the lowered PSA levels, and the overall well being. From what I take out of them, there is no worry with taking it long term even. Because it works a bit differently that Viagra, it doesn't "self activate" in that it doesn't force an erection on (when used for this purpose. I don't). Cialis actually just helps maintain an erection or makes it fuller. I would like to see an actual study on it that is fairly long term, but for now, I don't mind being the guinea pig, lol. I have nothing but good to say about it. Sometimes if I take it on an empty stomach, I get a little indigestion, but not always. I am all for it based on my own experiences, as well as a bunch of other men aged 23-60 that I know take it daily. They are all very happy with the overall effects.

I take 10mg a day. Some take this in two doses but its more convenient for me to just take it once a day, usually about 2-4 hours before bedtime . All cialis commercials state that its approved for daily use. I love it. The only couple of downsides is that for the first couple of weeks you may notice lower back aches and "delayed back pain" is also a side that's given on the commercials. The other downside, as Warmouth mentioned, is heartburn...horrendous heartburn, at least for me. But I love the stuff. I did experience the back ache as well, down into my hamstrings too. But that went away. The added vascularity is a nice bonus too.

Ha ha. Just seen this. We need to continue this. Perhaps I will
post an article I recently found. It is MUCH MORE potent than visage, and if you take 20mgs or 30mgs, it will last all flipping weekend for sure! I get effects if I miss a day at 10mgs.

there are no direct studies that I know of related to what you are describing however, many of my patients explain the same "effects" you are describing. tadalafil is a 36 hr drug... and I do know of these "wanted " side effects are being studied for other uses. many drugs have the approved FDA use but they have may have other uses that they can be prescribed for. at a EOD low dose use for those afore mentioned effects, just be careful as there is no specific research data to show any long term effects. I personally always keep tadalafil on hand but do not use it often...I do believe there is research stating it does have a positive effect on test though...

Cialis contains Tadalafil not Sildenafil like Viagra . Does Tadafil also effects testesterone levels ?
